The fastest way would be to take the Air Train from JFK airport to the end of the line in the Jamaica LI RR railroad station. The air train costs $5.00.
Once you get to the LI RR (Long Island Railroad Station) you can take an LI RR train to Penn Station in Manhattan (last stop).
Once in Penn Station in Manhattan you can take the subway. Take the IRT 7th ave line.
Get on the downtown local #1 subway line and take it to the last stop in Manhattan which is South Ferry.
Once you get there go up to the street and cross the street and enter the Ferry Terminal and catch the next Ferry to Staten Island. The Ferry is a 25 minute ride to Saint George Ferry Terminal in Staten Island. (the ferry boat ride from Manhattan to Staten Island is free)
You have now arrived at Saint George Ferry Terminal in Staten Island
1 Air train from Kennedy airport is $5.00
2 Long Island Railroad train from Jamaica to Penn station NY is off peak fare is $5.75
3- Subway ride is fare is $2.25
4- Ferry boat ride is free

Here’s how to get from JFK to the St. George Ferry Terminal in Staten Island.
Step 1: take the Airtrain from the terminal to Jamaica Station. Please listen for the announcement "Station D: Jamaica Station."
Step 2: at Jamaica Station, purchase a $7.25 MetroCard from the MetroCard Vending Machine, and exit the Airtrain system into Jamaica Station. The $5 fare will be deducted from the MetroCard, when exiting the Airtrain system at Jamaica Station. The remaining $2.25 will be use on the subway.
Step 3: Once thought turnstile (faregate), follow the directional signs to the Long Island Railroad. Purchase your LIRR ticket from the ticket machines on the pedestrian bridge overlooking the platform. A one way off peak ticket from Jamaica Station to Penn Station will cost you $5.75. If you’re arriving on a weekends, you can purchase a CityTicket for $3.50.
Once you purchase your ticket, look at the display board on the pedestrian bridge for the next arriving train bound for Penn Station. It will tell what track the next westbound train will arrive on, which is usually tracks 1, 2 or 3.
Step 4: at Penn Station, follow the directional sign to the New York City subway. Take the the downtown (southbound) #1 line (Red Line on the subway map) from Penn Station to South Ferry.
Step 5: at South Ferry, exit the subway into the Staten Island Ferry Terminal and take the Ferry from Lower Manhattan to Staten Island. The ride on the ferry is free.

Best route by subway is:
Step 1: Take AirTrain Howard Beach to Howard Beach Station.
\Buy $7.25 Regular/Pay-Per-Ride MetroCard to exit into subway mezz or paid $5 to exit area to purchase Unlimited MetroCard for subways. Subways and buses are $2.25
Step 2: Take Manhattan-bound (A) express to Broadway Nassau.
Step 3: Transfer to Brooklyn-bound (4)(5) two stops to Bowling Green.
Step 4: Follow the direction of train you were on and exit toward sign that read STATEN ISLAND FERRY.
Step 5: About few minutes walk, you’ll be in Whitehall Ferry Terminal (South Ferrry) where you could get on free Staten Island Ferry to St. George.
You could also use MetroCard on Staten Island Railway at St. George and Tompksville Station.
